La caffeina ha un evidente effetto protettivo contro la crescita delle cellule di melanoma umano. E’ la conclusione alla quale sono giunti i ricercatori dell’Istituto superiore di sanità in collaborazione con i colleghi degli Irccs Idi di Roma e Neuromed di Pozzilli e delle università di Ferrara e Tor Vergata di Roma, in uno studio appena pubblicato sulla rivista internazionale ‘Molecules’. Lo studio – si legge sul sito dell’Iss – è stato focalizzato sull’identificazione dei meccanismi attraverso i quali la caffeina svolge un importante ruolo protettivo contro alcuni tipi di tumori, già descritto in molti lavori in letteratura, ma ancora non completamente caratterizzato a livello molecolare. “Utilizzando approcci in silico e in vitro, abbiamo identificato una proteina che probabilmente – spiega Francesco Facchiano, coordinatore dello studio effettuato presso il Dipartimento di oncologia e medicina molecolare dell’Iss – gioca un ruolo fondamentale in questa azione benefica della caffeina, cioè l’enzima tirosinasi che, come è noto, ha una funzione chiave nella sintesi della melanina e che svolgerebbe sia un’azione protettiva contro gli effetti del danno generato dai raggi Uv, sia un’importante funzione di immunomodulazione. Infatti la melanina prodotta da cellule di melanoma umano esposte alla caffeina è risultata significativamente aumentata”. “Molto importante – sottolinea Claudio Tabolacci, primo autore dell’articolo e ricercatore sostenuto dalla Fondazione Umberto Veronesi – è stata la scelta dei modelli cellulari, che in questo studio sono le ‘melanoma initiating cells’ che hanno interessanti caratteristiche di staminalità, tra le quali la capacità di conferire resistenza ai farmaci e la recidiva di un tumore: la caffeina ha significativamente ridotto la crescita di queste cellule. Abbiamo inoltre evidenziato il ruolo di molecole di segnale come Il-1β, Ip-10, Mip-1α, Mip-1β e Rantes, la cui secrezione da parte di queste cellule in coltura è ridotta quando vengono esposte alla caffeina”. Come tutti i farmaci, anche la caffeina ha potenziali effetti collaterali, ma i risultati dello studio appena pubblicato aprono nuove ed interessanti prospettive nell’ambito della terapia differenziativa, finalizzata cioè a far differenziare le cellule per colpire solo quelle tumorali evitando la comparsa di recidive dopo il trattamento chemioterapico. Terapia che – conclude l’Iss – per tumori maligni come il melanoma cutaneo è considerata un promettente campo di studio.
