22, settembre 2023 – Scoperte mutazioni in 11 geni associate a forme aggressive di cancro alla prostata: è il risultato di uno studio di un team di ricerca guidato da scienziati del Center for Genetic Epidemiology della Keck School of Medicine dell’USC e dell’USC Norris Comprehensive Cancer Center ha. Pubblicato sulla rivista Jama Oncology, è il più grande studio sul cancro alla prostata che abbia mai esplorato l’esoma, cioè la parte del Dna che contiene le istruzioni per creare le proteine. Gli scienziati hanno analizzato i campioni di circa 17.500 pazienti affetti da cancro alla prostata. Il team ha analizzato campioni di sangue di pazienti con tumore alla prostata di origine europea, di cui 9.185 avevano una forma aggressiva della malattia e 8.361 no. Hanno confrontato la frequenza delle mutazioni tra i due gruppi. Nella prima fase delle indagini, hanno sequenziato l’intero insieme di geni codificanti per le proteine tra quasi un terzo dei partecipanti. Nella seconda fase, i ricercatori hanno utilizzato campioni provenienti dai partecipanti rimanenti per concentrarsi su un sottoinsieme di 1.749 geni che erano stati precedentemente associati al cancro o che erano emersi come probabili candidati nella prima fase. Gli undici geni emersi come portatori di mutazioni significativamente legate al cancro alla prostata aggressivo includono BRCA2, noto anche per la sua connessione con il cancro al seno. I risultati possono aiutare a personalizzare le cure. Lo studio ha scoperto mutazioni associate a un rischio maggiore di tumore alla prostata più aggressivo e letale che attualmente non sono incluse nei test genetici già in uso clinico. I ricercatori hanno anche scoperto che alcuni geni che fanno attualmente parte di test in uso non erano stati correlati in realtà prima d’ora al rischio di malattia aggressiva. “I risultati non sono completamente definitivi – sottolinea l’autore principale Christopher Haiman – c’è bisogno di fare ancora molto lavoro per determinare su quali geni gli oncologi dovrebbero concentrarsi nei test”.
